　故事在这个美丽的城市里会展开了一场反恐大战。得到上级命令，彩虹小队(Team Rainbow)火速赶到这里，发现原本平静的生活被国际恐怖组织破坏得七零八落，造成大量居民流离失所，无家可归。宽阔的街道上，随处都可看到被损坏的豪华车辆。作战中，反恐小组队长Logan Keller将带领他的队员同恐怖分子展开殊死拼搏。它有两个新的助手，分别是来自墨西哥的Kan Akahashi和Gabriel Nowak。游戏开始，Keller独自率先闯进Strip，(Strip俱乐部是美国伟大的建筑之一，实际上也是世界上最出色的一个墓地，它设立在维加斯这座城市中，就像一座天堂一样让人们十分崇拜和神往。)这两名新助手起先主要是进行高难度训练，以便等待执行更加危险的反恐任务。
在《Rainbow Six: Vegas》这款游戏中，除了Strip这个区域之外，第二个作战地点是Calyspo。想要进入这里执行任务，前提需要解决在墨西哥任务。队长Keller快速来到街道上，在那里会碰到Michael Walter和Jung Park两个角色，任务目的是营救“北大西洋公约组织(NATO)”的顾问“菲利普 史密斯(Philip Smythe)”先生。不管在那个区域，反恐小组必须速战，只有赶在恐怖组织头目Irena Morales可怕的计划之前粉碎他们的行动，才能顺利完成保护任务。
讲求战术，蛮乾等于失败!
　新作提供了一个崭新的彩虹六号游戏系统，进入Strip这个区域，你将会注意到Ubisoft开放了一个自由世界。作战环境变的十分巨大，玩家可以随心所以地率领自己的作战小组在街道上展开战斗，很多物体可以用来作掩护。在这里你也可以发现很多作战武器，如果需要的话也可以进行交换，比如在大环境中就不适合使用来复枪。彩虹小队起初使用的武器有冲锋枪，来复枪，手枪和其他各种轻武器等。很显然，像苏式武器AK-47在这里已经淘汰使用。
完全通过Strip这个区域后，便可以率领作战小组进入Casino之地，这里也被大量恐怖分子所占领，如何拿下他们确实需要精心布置一番。首先，你会发现自己所处的位置是在Casino地板上，要想解决目前的阻力，只有混合使用手榴弹和炮火来作战。维加斯这座城市具有一个可破坏的环境场所，使用武器轰炸和射击后会看到非常逼真的残损状况。Casino最难进攻的地方是旅馆，因为“菲利普 史密斯”先生被囚禁在那里，不能贸然行事否则任务马上就会失败。只有悄悄从屋顶着手，把人质安全救出之后方可实行大规模进攻。
